Padmanabhapuram Assembly constituency comprises the following areas of Kanniyakumari district in the South region of Tamil Nadu: Kalkulam Taluk (Part) Veerapuli R.F., Veerapuli Extn. (Kulasekharam), Thirparappu, Aruvikkarai, Churulacode, Velimalai Forest, Mecode, Attoor, Thiruvithancode, Thuckalay, Kalkulam, and Veeyanoor villages. Thiruparappu (TP), Thiruvattar (TP), Kulasekharam (TP), Ponmanai (TP), Kumarapuram (TP), Kothanalloor (TP), Verkilambi (TP), Attoor (TP), Padmanabhapuram (M), Thiruvithancode (TP) and Vilavur (TP).

This constituency covers an approximate area of 490 square kilometers and the geographic coordinates of Padmanabhapuram is: 8°23'04.9"N 77°20'38.0"E.

In the 2021 Tamil Nadu Assembly elections, Mano Thangaraj T of DMK won in this constituency beating John Thankam D of AIADMK by a margin of 26885 votes which was 15.9% of the total votes cast. In 2016, Mano Thangaraj T of DMK was the winner in this seat beating Rajendra Prasad K P of AIADMK by a margin of 40905 votes which was 25.32% of the total votes polled in the constituency. In the 2011 Assembly elections, Dr.Pushpa Leela Alban of DMK won this seat defeating S.Austin of DMDK by a margin of 19321 votes which was 13.38% of the total votes polled for the seat.
