Pudukkottai Assembly constituency comprises the following areas of Pudukkottai district in the Kaveri Delta region of Tamil Nadu: Pudukkottai Taluk Alangudi Taluk (Part) Pallavarayanpathai, IIaikadividuthi, Thirumanancheri, Pattathikadu, Kurumbivayal, Keelatheru, Therkutheru, Vadatheru, Vanakkankadu, Mullankuruchi South, Mullankuruchi North, Kanakkankadu, Karuppattipatti, Ayippatti, Valangondanviduthy, Vellalaviduthy, Adiranviduthy, Malaiyur, Therkutheru, Theethanipatty, Ponnamviduthy, Mankottai and Kalapam villages.

This constituency covers an approximate area of 505 square kilometers and the geographic coordinates of Pudukkottai is: 10°26'57.8"N 78°57'35.3"E.

In the 2021 Tamil Nadu Assembly elections, V Muthuraja of DMK won in this constituency beating Karthik Thondaiman of AIADMK by a margin of 13001 votes which was 7.25% of the total votes cast. In 2016, Periyannan Arassu of DMK was the winner in this seat beating Karthik Thondaiman of AIADMK by a margin of 2084 votes which was 1.22% of the total votes polled in the constituency. In the 2011 Assembly elections, Muthukumaran.P of CPI won this seat defeating Periyannan Arassu of DMK by a margin of 3101 votes which was 2.22% of the total votes polled for the seat.
