Vilathikulam Assembly constituency comprises the following areas of Thoothukkudi district in the South region of Tamil Nadu: Vilathikulam Taluk Ettayapuram Taluk Ottapidaram Taluk (Part) Kudiraikulam, Nagampatti, Passuvandanai, Muthuramalingapuram, Minakshipuram, Kumara Ettaiyapuram, Eppodumvendran, Kattunayakkanpatti, Adanur, Mullur, Muthukumarapuram, Veppalodai, Terku, Kalmedu, Vadanatham, Kolamparambu, Chandragiri, Jagaveerapandiapuram, K.Dalavaipuram, Vellaram, P.Duraiswamipuram, Kila Mudiman, Keela Mangalam, Sillangulam, S.Kumarapuram, K.Shanmukapuram, D.Duraiswamipuram and Pattanamarudur villages.

In the 2021 Tamil Nadu Assembly elections, Markandayan V of DMK won in this constituency beating Chinnappan P of AIADMK by a margin of 38549 votes which was 23.2% of the total votes cast. In 2016, Uma Maheswari K of AIADMK was the winner in this seat beating Beemaraj S of DMK by a margin of 18718 votes which was 12.25% of the total votes polled in the constituency. In the 2011 Assembly elections, Markandayan. V of AIADMK won this seat defeating Perumalsamy. K of INC by a margin of 22597 votes which was 16.95% of the total votes polled for the seat.
